Hi can I drill more than one hole in pinto thermostat

You can drill as many as you like if it's your thermostat 

In my 2B's pinto I used to drill one hole of about 3 - 4mm, positioned so that it would be near the top of the thermostat when it was fitted.  I found that this was enough to prevent air locks without defeating the operation of the stat.
